Context
Cryogrinding or Cryogenic grinding is an efficient method for mechanic tissue disruption. With cryogrinding using Precellys, solid and resilient tissue samples (e.g. heart and skeletal muscle, bone tissue) can be rendered into powder in only a few seconds (a great benefit of this technique, compared to the time consuming manual grinding with mortar and pestle). Another essential advantage of cryogrinding is the homogenization of tissues at low temperature (0 – 10 ° C) thus preventing the chemical transformation of unstable metabolites, such as specific classes of signaling lipids and energy currency metabolites. Following the cryogrinding, the tissue powder can be easily handled (e.g. aliquoted, weighted) which facilitates further polar and lipid metabolite extraction using different analytical methods.
Here, we demonstrate how to perform cryogrinding using Precellys® Evolution, Cryolys® Evolution, and metal tubes.
